Laragen is the first commercial company to apply real-time PCR technique in transgenic genotyping. In 2001, Laragen successfully applied Taqman technique to genotype APP and PS1 double transgenic mice.

Laragen developed a proprietary high throughput DNA extraction technique to extract high quality genomic DNA from mouse/rat tissues. High quality genomic DNA, robust PCR assays and stringent QC makes our mouse genotyping results very accurate (>99.95%).

For sample preparation, we require 0.3 to 0.4 cm tail clips and 2 mm ear punches. It is important that the tissue samples have similar sizes. Samples can be sent in either 1.7 ml Eppendorf tubes or 1.1ml 8-strip microtubes in 96-well boxes.

Samples in 96-well plate format should be arranged by the columns as indicated below:

Depending on the number of samples and nature of the tests, we either waive the initial set-up fee or only charge a nominal set-up fee to cover the reagent cost. For the mouse models that are not in the Laragen test pool, we need to have the following to develop and convert to the Laragen mouse genotyping protocols:

  • JAX strain number
  • Existing mouse genotyping protocols
  • Transgene construct sequences
  • Transgenic integration site or knock-in/knock-out sites if possible
  • Control samples to validate the tests

Probe Based TaqMan Mouse Genotyping Service

Most of Laragen’s mouse genotyping tests are probe-based TaqMan assays. Laragen’s TaqMan assays have an internal positive control on a housekeeping gene to ensure the test integrity. Also, all tests have CAR(carrier), HOMO and HET(if applicable), WT(wildtype) and NTC controls. The samples in the grey zone will be repeated multiple times to ensure test accuracy, free of charge.

Gel Based PCR Mouse Genotyping Service

A small portion of Laragen’s mouse genotyping tests are gel-based PCR assays. Like probe-based assays, we also include an internal positive control PCR on a house-keeping gene to ensure PCR integrity. All PCR reactions are optimized to yield accurate genotyping results even at suboptimal conditions. A vigorous SOP is implemented to eliminate PCR contamination problems.

Heterozygosity Mouse Genotyping by TaqMan Service

Laragen also provides heterozygosity tests for transgenic mice-based on qPCR (TaqMan) via copy number analysis. Normalized high quality genomic DNA samples are tested in triplicates and the transgene copy number of unknown samples are compared with the known het and homo controls.

SNP Genotyping Service

Laragen offers low to medium throughput SNP genotyping by the following two platforms:

  • SNP genotyping by Sanger sequencing.
  • SNP genotyping by TaqMan allelic discrimination (AD)
  • SNP genotyping by MiSeq Genotyping by Sequencing (GBS)

Trinucleotide Repeat Sizing Analysis

Laragen has more than 10 years experience on trinucleotide repeat sizing analysis from various mouse models:

  • CAG repeat numbers in Huntington's diseases (HD), Spinocerebellar ataxia type 1 (SCA1), Spinocerebellar ataxia type 3 (SCA3), Spinocerebellar ataxia type 7 (SCA7), and Spinal and bulbar muscular atrophy (SBMA) mouse models
  • CTG repeat numbers in Myotonic dystrophy type 1 (DM1) mouse models
  • GAA repeat numbers in Friedreich's ataxia (FRDA) mouse models
