To download your sequences results, please login
in DNALIMS account at
sequencing.laragen.com
or 66.245.244.196.
The
old sequences at Laragen FTP server is still available to download.
You can download the old sequences with either a web browser or a ftp software
·
Downloading with a Web
Browser:
To download your sequences using any
browser, type your Client ID number and our server information into your browser's
address window in the following format:
ftp://clientid@66.245.244.196
After typing this into your browsers
address window you will be prompted to supply a password. Supplying the
correct password will take you to your home directory where your files reside.
For example, your can download the
some sample file from our demo folder by using the following client ID and
password:
Client ID = dem100
Password = demo
Type the following into your browser
and press return:
ftp://dem100@66.245.244.196
in the password window, type demo and press return. You will see a
folder called demo. Double click the folder will lead you to a file call
sequence.sit. You can download the file by double clicking it.
·
Downloading with an FTP
program:
For MacIntosh 9.2 or earlier, we
recommend using the Fetch program
to download your sequences. For Windows user, we recommend using FTP Explorer or WS FTP LE. You should NOT use
passive mode transfers (PASV) with these FTP programs. The following is the
default setting for Fetch (under Customerize -> Preference -> Firewall
-> Use passive mode transfers (PASV)). You do not need to change anything
when using FETCH. However, you need to change the settings in both FTP Explorer
and WS FTP. With FTP Explorer, this is under View -> Options -> Fire Wall
-> Use PASV mode. With WS FTP, this is under Option -> Session -> Use
Passive Transfer Mode. Please Remove checks from all these settings.
To download your sequences, please
use the following settings in your FTP programs:
Host Name : FTP.LARAGEN.COM
User Name : your client ID
Password : your password.
·
Uncompressing zip files
after downloading:
You need either WinZip or Stuffit
Expander to unzip the DNA sequence file compressed in zip format. A
older version of Stuffit
Expander for Windows can be downloaded from our server.
·
Converting trace files to
MacIntosh Classic Format:
If you are still a classic MacIntosh
user, you need to convert the trace file (.ab1) to classic Mac format
(MacOS 9.2 or earlier). You also need ABI File Type Converter to
convert the electropherograph files from PC/Unix format to classic MacIntosh
format.
·
Viewing the sequence trace
files:
Each sequencing result comes with
two files. One is plain text file (.seq). The other is electropherograph file
(.ab1). You can open the .seq file with any word processing or text editor.
However we strongly recommend that you should view electropherograph file. For
MacIntosh Computer, you can use EditView from Applied
Biosytems or FinchTV
from Geospiza. Both EditView and FinchTV are freeware. For Window
based PC, you can use Chromas.
The older version 1.45 is freeware. FinchTV also has a Window version.
However, my favorite sequence viewer is Sequence
Scanner from Applied Biosystems. This free software provides quality
scores and ability to scale the signal intensity.
